Training objectives
- The course represents the first teaching of pharmaceutical analysis with laboratory exercises to individual places applied to the quantitative determination of biologically active substances. The goal of the course is to provide students with the knowledge of the main analytical methods applied to the quantitative determination and dosage of drugs with practical procedures that provide for the treatment of the sample, the choice of the analytical procedure, the analysis and processing of experimental data obtained. The course aims to give students the ability to act in safety and accuracy in the laboratory in performing quantitative analysis of active ingredients and to report analytically on the determinations made in the laboratory. Students will have the opportunity to use the main and common analytical instruments applied quantitative chemical analysis.
The theoretical part of the course will acquire knowledge on the concepts and laws that are the basis of the main techniques and analytical methods and their applications according to the chemical and physical properties of the analytes.
The practical part will apply the theoretical knowledge acquired through laboratory exercises to individual places that provide the common methods of preparation of the sample containing the analyte, the quantitative analysis that can be done by volumetric or instrumental, and finally the statistical treatment of experimental data obtained. Prerequisites
- In order to understand and assimilate the course content it is essential to have acquired the knowledge provided by the courses: general chemistry, organic chemistry, analytical chemistry.
a)To know the main physical-chemical characteristics of inorganic compounds.
b)Structure, nomenclature and property of features and major organic molecules.
c)Basic concepts, characteristics and applications of the main analytical techniques. Course programme
- The course includes 5 credits equal to 40 hours of frontal teaching and 4 credits equal to 48 hours of laboratory exercises in single place.
Theoretical part.
Introduction concerning the main methods of quantitative analysis reported in the official pharmacopoeia for the dosage of the active ingredients. Equipment, materials and reagents in quantitative analysis. Statistical processing of data in analytical determinations, Physico-chemical properties of the analytes, Main extraction methods, Weight analysis (10 hours).
Introduction to volumetric analysis, Volumetric analysis: acid / base volumetric determinations (acidimetry, alkalimetry), Volumetric analysis: complexometry and chelometry, redox titrations, Volumetric analysis: titrations in non-aqueous solvents, titrations with precipitate formation, Volumetric analysis: potentiometric titrations and conductivity meters (18 hours).
Chromatographic methods of analysis: main chromatographic techniques TLC, HPLC, gas chromatography; column theory and quantitative determinations, Optical methods of analysis: generalities and basic theoretical principles, quantitative analysis by UV-visible spectroscopy, Optical methods of analysis: atomic spectroscopy (emission and absorption) and polarimetry (12 hours).
Laboratory exercises.
Acidimetric titrations (12 hours). Alkalimetric titrations (12 hours). Chelometric titrations (6 hours). Permanganometric titrations (6 hours). Iodometric titrations (3 hours). Conductimetric Titrations (3 hours). Quantitative determination of UV-visible active ingredients (6 hours). Didactic methods
- The course includes:
a)Classroom lectures IN PRESENCE on topics related to the theoretical part of the course (40 hours).
b)Exercises to individual places in the laboratory teaching concerning the quantitative analysis of biologically active substances by means of volumetric methods (36 hours) and instrumental determinations (12 hours). Learning assessment procedures
- The achievement of the training objectives set by the course is verified by means of an oral exam which includes:
Verification of learning regarding the topics proposed by the course; in particular, the choice of extraction and quantitative analytical methods according to the chemical-physical characteristics of the compounds under examination, theoretical bases and applications of the methods and analysis of the experimental data obtained. The evaluation will take into account the laboratory experiences and errors obtained in the individual tests. Reference texts
