Training objectives
- Formative purposes:
- Cognitives: knowledge of the most important analytical methods in the qualitative determinations of the drugs, knowledge about how to handle the drugs and how to choose the right conditions to proceed in the analysis and in the treatment of the data.
- Operatives: Be able to handle the drugs in conditions of safety, capacity to give short communications about the data obtained. Be able to use properly the principal instrumentations for the analytical chemistry.
- Conducts: Be able to choose the most suitable analytical methods according with the sample to analyse, be able to describe the data obtained in a critical way, proposing an explanation about the fenomeno observed. Prerequisites
- Basic principles of Inorganic Chemistry, Organic Chemistry, Analytical Chemistry, Medicinal Chemistry
Course programme
- Contents: Theoretical part: Introduction about the most utilized methods for the qualitative analysis following the indications reported in the Farmacopea Ufficiale Italiana XII edizione and European Pharmacopoeia 10th Edition. General notices. Monographs: Titles; Related atomic and molecular masses; Definition; Characters (solubility); Identification (first and second identification); Tests and assays (scope, calculation, limits, indication of permitted of impurities); reference substances, reference spectra. Physical and physicochemical methods: optical rotation, melting point, atomic absorption spectrometry, absorption spectrophotometry, infrared, ultraviolet, visibile, thin layer chromatography. Identification reactions of ions and functional groups. Limit tests. Reagents. Laboratory practice. Identification of drugs following the indications reported in the Farmacopea Ufficiale Italiana XII edizione and European Pharmacopoeia 8th Edition. Identification reactions of ions and functional groups. Identification by physical and physicochemical methods. Isolation of drugs from complex mixtures.
Didactic methods
- The course will be delivered through theoretical lessons aimed at exposing and explaining the contents of the program, integrated with practical examples, case studies, exercises and moments of study.
Learning assessment procedures
- Oral exam: (maximum time of 30 minutes) consists of a theoretical identification of a random substance chosen from the U.F., a question about e.g. IR, UV, Polarimetry, Solubility, Melting Point, etc and the comment of a reaction mechanism that is the basis of a colorimetric assay.
The final evaluation may range from 18 (sufficient response to all three questions) to 33 (optimal preparation). The final grade will also depend on the laboratory tests. The professor and tutors will evaluate the student's behavior, commitment and approach to the identification of unknown substances. Reference texts
